A Multi-Omic Framework for Understanding KRAS Inhibitor Response

A Champions White Paper

KRAS-mutant tumors remain among the most biologically complex and clinically challenging cancers to treat. After decades in which KRAS was considered undruggable, the emergence of allele-specific inhibitors and, more recently, pan-RAS(ON) inhibitors, has created new therapeutic opportunities across non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and pancreatic ductal adenocarcinoma (PDAC).

Champions Oncology has applied a clinically relevant platform combining patient-derived xenograft (PDX) models with integrated multi-omic profiling. A KRAS-mutant PDX cohort spanning lung, colorectal, and pancreatic cancers has been used to investigate sensitivity and resistance to pan-RAS(ON) inhibition.

Champions Oncology’s integrated platform provides exactly this foundation, offering drug developers a clinically grounded, mechanistically informed path to understanding and ultimately outpacing resistance in KRAS-mutant disease.
